  • The Medium of the Culture Tissue?

    Hi everyone,

    I am newbie here.

    I do not know exactly which one is the true term, the culture tissue or the tissue culture.

    I would like to ask a question.

    What is the medium (liquid?) is used in the tissue culture method such as below image?

        Well, I haven't looked at the link (waiting for the OP to clarify what they're up to), but if it's agar, I've got vast experience: I've made and used many tens of thousands of such culture media in the years when I worked as an infectious diseases scientist.

              So, if I would like to try the culture tissue method, all I have to do is just buy the agar then immerse the seed in it?

                Surely if you are doing a tissue culture you are growing a new plant from existing plant material - not a seed at all?

                    The agar will require various nutrients, which would normally be supplied by the roots.

                    It'll probably also require some antibiotics and antifungals (we used to use Fluconazol, Ceftazidime and Vancomycin, although some organisms now have evolved resistance to these) to prevent micro-organisms growing in or on it.
                    If you're not careful and don't use anti-microbials, your nutrient-enriched agar might end up growing all manner of bacteria and fungi; including some unpleasant micro-organisms which could make you ill. Fungal spores can cause respiratory irritation or induce asthma attacks and there's even a small chance that you might end up accidentally culturing some Bacillus anthracis (ANTHRAX) which is a soil-borne organism, or Clostridium tetani (TETANUS).

                          I understand that some rootstocks - such as M116 - are propagated in this way, as it's quicker than stooling/layering if the rootstock is a bit slow to root.

                          (slow rooting and more difficult propagation is why M116 never found success until lots of the commercial MM106's began to die from root diseases; a replacement rootstock with better disease resistance was urgently needed and M116 was too slow to propagate in the traditional way).
