Yes you can cut a hole, but it is tricky and you might break a couple getting one done right.
In commercial units the panel with the hole is often metal for that bit and glass for the rest of the panel.
Why have the but inside in the first place? I'm another for having it outside and running a pipe inside, perhaps underground and with a tap inside (as well as out).
I know some people put black water butts in a greenhouse to absorb day time sun and regulate night time temps, but the space cost is huge.

We opted to have our 3 water butts inside our greenhouse.... but then it is quit big and we have used it to our advantage. We used Perspex for the middle bit where the drain tube connects the butt to the outside guttering. It has worked very well
If the panel is polycarb then you should have no problem, if not then replace the glass with a sheet of polycarb.
