Is it an old leaf? I expect it is just coming to the end of it's natural life. Take it off if it bothers you.

You can frighten yourself silly with blotches on tomato leaves - there is an identification key here: Tomato leaf symptoms Diagnostic Key

However, if you are growing in containers much the most likely cause is early signs of magnesium deficiency, so give them a feed of something like Tomorite.

I give mine a dose of Epsom salts, does wonders for their joints...

Thanks, it looks like I have magnesium deficiency. So going to give them a feed.
